Amphlett wuchs in Geelong, Victoria, Australien auf. Sie verließ ihr Zuhause als Teenager und reiste nach England, Frankreich und Spanien. Sie wurde drei Monate in Spanien festgehalten, weil sie auf der Straße gesungen hatte. Im Mai 2001 wurde Divinyls '"Science Fiction", geschrieben von Amphlett und Leadgitarrist Mark McEntee, von der Australasian Performing Rights Association als einer der 30 besten australischen Songs aller Zeiten ausgewählt.
