Mothersbaugh wurde Ende der 1970er Jahre als Mitbegründer, Leadsänger und Keyboarder der New-Wave-Band Devo bekannt, deren "Whip It" 1980 eine Top-20-Single in den USA war und die seither Kult bleibt. Mothersbaugh ist einer der Hauptkomponisten von Devos Musik. Neben seiner Arbeit mit Devo hat Mothersbaugh über seine Produktionsfirma Mutato Muzika Musik für Fernsehserien, Filme und Videospiele gemacht. Er komponierte die Musik für die 13-jährige Serie der Zeichentrickserie Rugrats und der drei dazugehörigen Kinofilme. Als Solomusiker hat Mothersbaugh vier Studioalben veröffentlicht: Muzik for Insomniaks, Muzik for the Gallery, Joyeux Mutato und The Most Powerful Healing Muzik in the Entire World. 2004 erhielt er den Richard Kirk Award bei den BMI Film and TV Awards für seine Beiträge zur Film- und Fernsehmusik.
